Laboratory Handling

Specimens collected at Auckland hospital: Specimens must be delivered to LabPLUS within 60 mins of collection. At the time of collection, place an "Urgent" label or Complement Pathways Activity sticker on the form. If only an urgent sticker is used, write clearly below it " Complement Pathways Activity " . Contact Immunochemistry on 09 3074949 ext 22113, Monday to Friday between 0800 and 1430 to inform the laboratory once the sample has been collected. Micro-collects are not suitable for pathway activity assays due to contamination with tissue fluids.

Transport

Ambient. Sample could arrive via Lamson and/or via the Front Counter.

Registration

Lamson staff: Ichem staff will complete a complement pathway assay tracking form (LPVIMPROFO986) which they will hand to you when a sample for complement pathway testing is due to arrive. When the sample is received, time stamp the request form and give the sample, request form and tracking form to the registration staff immediately for registration. Registration staff: Register the sample urgently and deliver the sample and the tracking form to an Ichem staff member. If no tracking form is received, register the sample urgently and hand deliver the sample to an Ichem staff member. After hours: If a sample is received after hours when no Ichem staff is available, receive the sample, give the sample to be registered urgently. The sample needs to stay at room temperature to clot for 60-90 mins. Centrifuge the sample at 3000 rpm for 5 mins. Separate the serum and store at -20°C in specimen services freezer (freezer 36 shelf 2 in the Ichem rack). Ichem staff will retrieve the sample on the next working day.

Separating

It is crucial the sample is clotted at room temperature for 60-90 mins before spinning and frozen immediately after spinning at -20°C
