Bone marrow aspirate is the preferred specimen type for analysis. Peripheral blood should only be submitted when bone marrow is unobtainable and there are sufficient circulating lymphoplasmacytic cells to enable reliable detection of the MYD88 c.794T>C (p.Leu265Pro) variant.

Test Method

This assay detects the MYD88 c.794T>C (p.Leu265Pro) variant by allele-specific PCR, with an analytical sensitivity of approximately 1.0% variant allele frequency. The assay is based on the method described by Varettoni et al. (Blood 2013;121:2522-2528). A negative result does not exclude the presence of a low-level clone below the limit of detection or a non-L265P MYD88 variant.
